Current Working Policy

Viewing your images via the FSU Appion website:

You will need to have an FSU Appion account created.  You will need to give Duncan:

  • A login name for your lab or, if you prefer, login names for each person you want to have separate access.
  • A short name for each project you want to use the Titan for (e.g., Actin Tropomyosin Hextorine Filaments).
  • Which login you want to own which projects.

Once your account(s) has been setup, you will be able to login and see images, etc. at:  https://myamiweb.rcc.fsu.edu/myamiweb_rcc

Getting Your Images:

When your data is collected two types of images are created.  The 1st type are regular images that are collected during Leginon operation and exposure imaging.  These images are summed images from raw frames.  The 2nd type are frames images that are only saved for exposure images and will make up the bulk of your data.  Once your data has been collected, your images (both regular and frames) will be transferred to the external hard drives you have sent us.  This process will take at least a week depending on how much data there is and may take longer.

 

What’s on Your Hard Drive:

Your hard drives will contain all your data as well as a metadata text file ([session_name]_metadata.txt).  Your data will consist of image data, frames data, references, and any processing data we’ve created. 

 

Image References:

When it comes to dark and bright references, each image is connected to a dark and bright reference (there may be more than one per project or for the screening data there may just be one set).  You can find the name of each reference in the metadata text file or on the website.  To find the data on the website:

  • click on an exposure image in one of your projects (an image that ends in 'ex')
  • click the 'i' button for image information (above the image and image details)
  • click the on the 'Data Tree' list to list the image details
  • click on 'dark' or ‘bright’
  • click on 'image' to get the image name

 

Direct Image Download from FSU

 

For In order to download images from FSU, you will need to create an FSU HPC account and setup file transfer protocols.

  • The  1st step is to setup and HPC account.  Follow the signup link (https://acct.rcc.fsu.edu/account/signup) and enter your your sponsor.
  • The 2nd step is to setup off campus access using a VPN client: (https://rcc.fsu.edu/doc/off-campus-vpn-access).
  • The 3rd step is to setup the Globus file transfer system (https://rcc.fsu.edu/docs/globus). 
  • The 4th step is to download your images.  Both your ‘leginondata’ and reference images will be located under the /lustre/cryo/lustre/leginondata/ directory.  Your frames data will be mailed to you as it is too big to download efficiently.
